Project 1:  Hand in hand Theory and experiments for the effective treatment of Parkinson’s disease.

What is Parkinson’s Disease?

Parkinson’s disease (PD) is the second most common neuro-degenerative disease and the number of patients with this Parkinson’s Disease is expected to increase over the next ten years.

The main treatment for PD is levodopa. Levodopa treatment is a combination of L-DOPA and decarboxylase inhibitors to ensure that levodopa is not converted peripherally by human dopa decarboxylase (DDC) before reaching the brain.

However, the bioavailability of levodopa needed to provide enough dopamine to reach the brain varies significantly in Parkinson’s patients.

In this interdisciplinary project, in close collaboration with experimental and theoretical research groups, human DDC and TDC bacteria in the presence of levodopa and a number of new inhibitors and classic carbidopa inhibitors were investigated to:

  1. Track changes in their mechanisms.
  2. Identify binding roles of the main amino acids involved in binding of the substrate and
  3. Determine the effect of mutations.

The new integration between theory and experiment will not only contribute to the creation of new inhibitors, but will also highlight interactions between theoretical and experimental research and illustrate the power of these combined efforts.
